Viable human embryos carrying a disease mutation have now been corrected by CRISPR-Cas9 editing. Although the correction is not perfect, and the embryos have not been implanted, this brings us closer to human germline editing and to the associated ethical questions.

Les cellules lymphoïdes innées (innate lymphoid cells, ILC) sont des effecteurs tissulaires qui se distinguent des lymphocytes B et T par l’absence de récepteur de l’antigène issu de réarrangements géniques. Elles se répartissent en deux groupes : les ILC cytotoxiques (NK) qui dépendent d’Eomes et de T-bet et produisent de l’IFN-γ et les ILC auxiliaires (ILC1, ILC2, ILC3) qui dépendent de T-bet, GATA-3 ou RORγT et produisent de l’IFN-γ, et de l’IL-5/IL-13 ou de l’IL-17/IL-22 respectivement. Récemment décrites, les ILC font l’objet d’un grand nombre de travaux les caractérisant comme étant parmi les premiers effecteurs de la réponse immunitaire innée mais aussi comme des acteurs du développement de nombreuses pathologies inflammatoires.; Innate Lymphoid Cells (ILC) are tissue-resident effector lymphocytes that differ from the B and T lymphocytes subsets since they lack specific antigen receptors derived from gene rearrangements. They can be divided into two groups, cytotoxic ILC (NK cells) which are dependent on the transcription factors Eomes and T-bet and produce IFN-γ, and helper-like ILC (ILC1, ILC2, ILC3) which are dependent on T-bet, GATA-3 or RORγT and produce IFN-γ, IL-5/IL-13 or IL-17/IL-22 respectively. Although they were described recently, ILC are the subject of a large number of studies that characterize their role as one of the first effectors of the innate immune response as well as in the development of a wide number of inflammatory disorders.

La grippe et ses complications restent un problème important de santé publique et une lourde préoccupation socio-économique. La découverte de nouveaux antiviraux et l’exploitation des connaissances sur l’immunité antivirale devraient offrir prochainement de nouvelles solutions thérapeutiques ou préventives pour mieux contrôler la sévérité de la grippe. Une meilleure compréhension de la pathogenèse devrait également aboutir à la découverte de molécules clés dont on pourrait moduler la production pour le bien-être des patients. Enfin, l’identification de nouveaux immunomodulateurs et immunostimulants pourrait permettre de compenser le dysfonctionnement des mécanismes de défense contre les infections bactériennes secondaires dont on sait qu’elles amplifient gravement le tableau clinique des patients grippés.; Influenza virus infection (“flu”) constitutes an important public health issue and is associated with a significant socio-economic burden. The discovery of new antivirals and a better understanding of antiviral immunity might offer new options for preventing or controlling flu infections. A better understanding of the underlying pathomechanism might also prompt the discovery of key molecules whose activity could be modulated in a clinical setting. Lastly, the development of new immunomodulators and immunostimulants might compensate for impaired host defense mechanisms and improve resistance to secondary bacterial infections, since the latter significantly worsen the clinical status of influenza-infected patients.
