dc.description.abstractLes progrès dans la connaissance du système des endothélines, puissants peptides vasomoteurs produits par l’endothélium vasculaire, ont été spectaculaires, depuis leur biosynthèse grâce à l’enzyme de conversion de l’endothéline (ECE), jusqu’à la pharmacologie de leurs récepteurs, ETA et ETB. La présence d’activités enzymatiques distinctes de l’ECE spécifiques de certains organes fait envisager le développement d’inhibiteurs sélectifs. Les récepteurs sur les cellules musculaires lisses vasculaires et sur les cellules endothéliales relaient, respectivement, des actions vasoconstrictrices (surtout ETA) et vasodilatatrices (essentiellement ETB). L’étude de la contribution des endothélines à la régulation du système cardiovasculaire et à l’étiologie de certaines affections vasculaires est l’objet d’une forte compétition car ces mises à jour pourraient conduire au développement d’agents thérapeutiques efficaces.fr
dc.description.abstractMultidisciplinary approaches including pharmacology, biochemistry, immunology and rapidly developing molecular biology techniques have been instrumental in the gigantic leaps observed in the field of endothelins in recent years. With the receptors cloned as well as proposed biosynthetic pathways being rapidly elucidated, the role of endothelin in cardiovascular homeostasis as well as in the ethiology of vascular pathologies may lead to the development of therapeutically useful agents. This review succinctly describes some of the important aspects of the biology of endothelins from the processing of pre-pro-endothelin-1 to the active moiety of endothelin-1. Furthermore, physiopathological considerations are presented concerning the usefulness of mixed ET(A)/ET(B) receptors as opposed to selective ET(A) receptors. Moreover, distinct endothelin-converting enzyme activity specific to particular organs is outlined. Finally, the nuclear and cytosolic calcium-mobilizing properties of ET-1 in the vascular smooth muscle are illustrated. As far as the contribution of endothelin in cardiovascular regulation is concerned, a correlation attempt will be made between the results obtained in animal models.en
