dc.description.abstract | variant hepatocyte nuclear
factor 1 (vHNF1) est un facteur
de transcription à homéodomaine
appartenant à une famille
hétérogène impliquée dans le
développement précoce et dans
le métabolisme. L’invalidation de
ce gène induit une létalité
embryonnaire précoce,
conséquence de l’absence
d’endoderme viscéral. Ce tissu,
d’origine extra-embryonnaire, est
indispensable pour la nutrition de
l’embryon, mais aussi pour la
régionalisation du neurectoderme
et la formation de la cavité proamniotique.
vHNF1 se place dans
une position prééminente
au sein du réseau complexe de
régulation contrôlant la
spécification et la différenciation
de l’endoderme viscéral. La
létalité embryonnaire précoce ne
permet pas d’étudier un éventuel
plus tardif rôle de vHNF1,
notamment pendant
l’organogenèse. Des mutations de
vHnf1 chez l’homme entraînent
des dysfonctionnements du
pancréas et des anomalies du
développement urogénital pour
l’étude desquelles d’autres
stratégies, seront nécessaires. | fr |
dc.description.abstract | Genetic and molecular evidence indicates that visceral endoderm, an extraembryonic lineage derived from primitive endoderm, is required for gastrulation, early anterior neural patterning, cell death and specification of posterior mesodermal cell fates. vHNF1, a homeodomain containing transcription factor belonging to the Hepatocyte Nuclear Factor (HNF) family, is first expressed in the primitive endoderm and is required for early embryogenesis and plays an essential role in visceral endoderm specification. Homozygous mutant embryos die just after implantation and before gastrulation displaying growth retardation, disorganised ectoderm and absence of visceral endoderm. Homozygous null blastocysts and differentiated embryonic stem cells do not express early or late visceral endoderm markers. Aggregation of vHnf1-deficient embryonic stem cells with wildtype tetraploid embryos rescues the periimplantation lethality and allows development to progress to early organogenesis, confirming that the early embryonic lethal phenotype is essentially due to defective extraebryonic tissue formation. These results, together with observations of other mutations affecting visceral endoderm function, in particular Hnf-4alpha, indicate that the specification of the visceral endoderm involves a complex regulatory network rather than a linear cascade. | en |
