L’initiation de la synthèse
protéique requiert la
reconnaissance du codon
d’initiation AUG d’un ARNm par
un ribosome portant l’ARN de
transfert initiateur chargé d’un
résidu méthionine (Met-ARNi
Met).
Chez les eucaryotes, l’initiation
s’opère en trois étapes
essentielles : la formation du
complexe de pré-initiation, sa
liaison à l’ARN messager, et enfin
son positionnement sur le codon
initiateur. L’intégrité du
mécanisme dépend d’une série
d’interactions moléculaires entre
les différents facteurs d’initiation,
le Met-ARNi
Met, la sous-unité
ribosomique 40S et l’ARNm.
L’initiation est un processus
complexe soumis à de multiples
contrôles. Ces mécanismes de
régulation sont exploités par
différents virus pour détourner
l’appareil traductionnel de la
cellule à leur profit. The initiation of protein synthesis requires the binding of a ribosome charged with the initiator methionyl-tRNA to the AUG initiation codon of an mRNA. In eukaryotes, initiation proceeds in three essential steps: the formation of the preinitiation complex, its recruitment to the mRNA and its migration to the correct initiator AUG. This process depends on a number of molecular interactions between the multiple initiation factors, the initiator Met-tRNA, the 40S ribosomal subunit and the mRNA. Translation initiation is under tight cellular control, which will often involve a modification of the interactions between initiation factors. Numerous viruses manipulate these regulatory mechanisms to usurp the translational machinery.
